Energy drink consumption has been anecdotally linked to the development of adverse cardiovascular effects in consumers, although clinical trials to support this link are lacking. The effects of Red Bull® energy drink on cardiovascular and neurologic functions were examined in college-aged students enrolled at Winona State University. In a double-blind experiment where normal calorie and low calorie Red Bull® were compared to normal and low calorie placebos, no changes in overall cardiovascular function nor blood glucose (mg/dL) were recorded in any participant (n = 68) throughout a 2-h test period. However, in the second experiment, nine male and twelve female participants subjected to a cold pressor test (CPT) before and after Red Bull® consumption showed a significant increase in blood sugar levels pre- and post Red Bull® consumption. There was a significant increase in diastolic blood pressure of the male volunteers immediately after submersion of the hand in the 5°C water for the CPT. Under the influence of Red Bull®, the increase in diastolic pressure for the male participants during the CPT was negated. There were no significant changes in the blood pressure of the female participants for the CPT with or without Red Bull®. Finally, the CPT was used to evaluate pain threshold and pain tolerance before and after Red Bull® consumption. Red Bull® consumption was associated with a significant increase in pain tolerance in all participants. These findings suggest that Red Bull® consumption ameliorates changes in blood pressure during stressful experiences and increases the participants’ pain tolerance.  相似文献

Bacillus subtilis microbe is commonly found in soil and produces proteases on nitrogen and carbon-containing sources and increases the fertility rate by degrading nitrogenous organic materials. The present study was aimed to develop hyper producing mutant strain of B. subtilis for the production of proteases, to improve the process variables by the response surface methodology (RSM) under central composite design (CCD) and the production of protease by the particular mutant strain in a liquid state fermentation media. The mutation of the strain was carried out using ethidium bromide. Pure B. subtilis strain was collected and screened for hyper-production of protease. The production of protease by mutant B. subtilis strain was optimized by varying temperature, inoculum size, pH and incubation time under liquid state fermentation. The CCD model were found to be reliable with r2 of 0.999. The maximum enzyme activity of B. subtilis IBL-04 mutant with 3 mL/100 mL inoculum size, 72 h fermentation time, pH 8, and 45 °C temperature was developed with enzyme activity 631.09 U/mL, indicates 1–7-fold increase in enzyme activity than the parent strain having 82.32 U/mL activity. These characteristics render its potential use in industries for pharmaceutical and dairy formulation.  相似文献

Salinity is extremely hazardous to agriculture worldwide and its expanding constantly. Soil of almost 100 countries facing salinity problem including Pakistan. Cyperus laevigatus also act as salinity indicator species is a naturally adapted halophyte dispersed in subtropical regions of world. Six populations of C. laevigatus were collected from different saline habitats to evaluate adaptations regarding anatomical and physiological characteristics. C. laevigatus is perfectly adapted to harsh environmental conditions like dry barren soils, saline lakes, hyper-saline wetlands and salt marshes. Ecological success of this species is due to plasticity in physiological and anatomical characteristics to adapt variable environmental conditions. C. laevigatus is a halophyte, exhibited increased biomass production in moderately saline habitat. Higher uptake of K+ occurs to compensate the uptake of Na+ ion contents, a striking feature of salt-tolerant and halophytic species. Accumulation of osmoprotectants like proline, free amino acids, soluble sugar and protein contribute significantly to osmotic adjustment. Stem thickness enhanced as salinity level of habitat increased to store water in parenchymatous tissues under physiological drought. Intensive sclerification in root cortex provide mechanical strength to plant as well as prevent the radial leakage of water. Well-developed aerenchyma, increased vascular bundle area, broader vessels, small and dense stomata are critical to cope with environmental hazards. Population of Jahlar lake showing maximum biomass production indicate that this species grows better in moderate salinities. Therefore, this species will prove very useful for revegetation of salt affected rangeland and prairies by direct growth of such halophytic ecotypes.  相似文献

In our present investigation, the crude methanol extract and chloroform fraction of the whole plant of Physalis minima Linn (Solanaceae) was investigated for anti-inflammatory, analgesic and antipyretic activities in NMRI mice and Wistar rats of either sex at 200 and 400 mg/kg, respectively. Various established in-vivo model's were used during the study. Both crude extract and chloroform fraction showed marked anti-inflammatory and analgesic activities as compared to a control at tested doses. The antipyretic potential of the crude extract and chloroform were insignificant in the Brewer's yeast fever model. Therefore, the whole plant of Physalis minima Linn could be considered as a potential candidate for bioactivity-guided isolation of natural anti-inflammatory and analgesic agents.  相似文献

The proteomic profile of hypothalamus, a key organ of CNS, is explored here by employing two widely used MS techniques, i.e. HPLC/ESI‐ion trap and HPLC/ESI‐quadrupole‐TOF MS. Strong cation exchange is used for the fractionation of peptides and protein search engine MASCOT is employed for data query. One hundred and thirty six proteins with 10 973 peptides were identified by HPLC/ESI‐ion trap MS, while 140 proteins with 32 183 peptides were characterized by HPLC/ESI‐quadrupole‐TOF MS. Among the total 198 proteins identified in both experiments, 78 proteins were common in both sets of conditions. The rest of the 120 proteins were identified distinctly in both MS strategies, i.e. 58 unique proteins were found using the quadrupole‐TOF while 62 were found with the HPLC/ESI‐ion trap. Moreover, these proteins were classified into groups based on their functions performed in the body. Results presented here identified some important signal and cellular defense proteins inevitable for survival in stressed conditions. Additionally, it is also shown that any single MS strategy is not reliable for good results due to loss of data depending on sensitivity of the instrument used.  相似文献

Background

Many drug development and toxicology studies are performed using cells grown in monolayers in well-plates and flasks, despite the fact that these are widely held to be different to cells found in the native environment. 3D, tissue engineered, organotypical tissue culture systems have been developed to be more representative of the native tissue environment than standard monolayer cultures. Whilst the biochemical differences between cells grown in 2D and 3D culture have been explored, the changes on the electrophysiological properties of the cells have not.

Methods

We compared the electrophysiological properties of primary normal oral keratinocytes (nOK) and cancerous abnormal oral keratinocytes (aOK), cultured in standard monolayer and reconstituted 3D organotypical tissue cultures. The electrophysiological properties of populations of the cells were analysed using dielectrophoresis. The intracellular conductivity of aOK was significantly increased when grown in organotypical cultures compared to counterpart cells grown in monolayer cultures.

Results

3D cultured aOK showed almost identical intracellular conductivity to nOK also grown in organotypical cultures, but significantly different to aOK grown in monolayers. The effective membrane capacitance of aOK grown in 3D was found to be significantly higher than nOK, but there was no significant difference between the electrophysiological properties of nOK grown in 2D and 3D cultures.

General significance

This work suggests that factors such as cell shape and cytoplasmic trafficking between cells play an important role in their electrophysiology, and highlights the need to use in vitro models more representative of native tissue when studying cell electrophysiological properties.  相似文献

The first purpose of our study was to determine culturable indoor airborne fungi in different places in Ahvaz during normal and dust event days. Ahvaz is the biggest city in southwest of Iran which has suffered from dust storms in recent decade. To minimize personal exposure to polluted air, governmental and private sectors excluding critical places such as hospitals, fire department and security organization are closed during dust event days. Therefore, the second objective of this survey was to see whether this plan (forced shutdown) is in place. Samples were collected from July 2010 to March 2011 by using quick take 30 equipped with a single-stage sampler. Fungal population was measured in school, university and hospital during normal and dust event days. Five dominant fungi genera in three sampling stations were Cladosporium, Alternaria, Aspergillus, Penicillium and Rhizopus the concentrations (CFU/m3) of which were higher during dust events than normal days. Airborne fungi such as Aspergillus, Mucor and Rhizopus are opportunistic fungi which can cause aspergillusis and mucormycosis infections in immunodeficiency individuals, vulnerable groups as well as healthy people. The results of this study confirmed the measure of forced shutdown in Ahvaz city due to increased concentration of fungi during dust event days.  相似文献

An increasing body of evidence suggested that intracellular lipid metabolism is dramatically perturbed in various cardiovascular and neurodegenerative diseases with genetic and lifestyle components (e.g., dietary factors). Therefore, a lipidomic approach was also developed to suggest possible mechanisms underlying Alzheimer’s disease (AD). Neural membranes contain several classes of glycerophospholipids (GPs), that not only constitute their backbone but also provide the membrane with a suitable environment, fluidity, and ion permeability. In this review article, we focused our attention on GP and GP-derived lipid mediators suggested to be involved in AD pathology. Degradation of GPs by phospholipase A2 can release two important brain polyunsaturated fatty acids (PUFAs), e.g., arachidonic acid and docosahexaenoic acid, linked together by a delicate equilibrium. Non-enzymatic and enzymatic oxidation of these PUFAs produces several lipid mediators, all closely associated with neuronal pathways involved in AD neurobiology, suggesting that an interplay among lipids occurs in brain tissue. In this complex GP meshwork, the search for a specific modulating enzyme able to shift the metabolic pathway towards a neuroprotective role as well as a better knowledge about how lipid dietary modulation may act to slow the neurodegenerative processes, represent an essential step to delay the onset of AD and its progression. Also, in this way it may be possible to suggest new preventive or therapeutic options that can beneficially modify the course of this devastating disease.  相似文献
